Chances are, it chose a drive letter that is conflicting with another drive.
Typically this happens in network situations when the admin assigns a new Mapped Network Drive.
Try this:
- right-click on "my computer" and left-click manage
- click "Disk Management" under "Storage"
- locate the drive that represents your thumb drive
- right click, select assign drive letter
- pick an unused drive letter
- click ok & you're done
